Antibacterial and Regenerative Testing

WP3 is dedicated to systematic in-vitro evaluation of the multifunctional MXene-based wound healing platforms, bridging material development and preclinical validation. Its primary objectives are to:

  • Assess biocompatibility and cytotoxicity of electrospun membranes loaded with MXene-PDA-Ab and MXene-PDA-HATMSC complexes using standardized ISO 10993-5 protocols, ensuring cell safety and viability.
  • Evaluate antibacterial performance of MXene-PDA-Ab conjugate–loaded membranes under near-infrared (NIR) photothermal therapy conditions, quantifying bacterial killing efficiency against MRSA and biofilm-disrupting capacity.
  • Investigate regenerative potential of MXene-PDA-HATMSC scaffolds by measuring their effects on fibroblast proliferation, keratinocyte migration (scratch assay), and endothelial cell tube formation, under both normoxic and hypoxic conditions to mimic chronic wounds.
  • Validate advanced in-vitro wound healing models that replicate the hostile microenvironment of chronic, infected wounds (hypoxia, nutrient deprivation, bacterial presence), establishing robust preclinical data prior to in-vivo testing. 

WP3 will generate critical evidence of therapeutic efficacy and safety, de-risking the transition to animal studies and accelerating the path toward clinical translation.
